Abstract

Kynurenine pathway is one of the important pathways in tryptophan metabolism. Its products include quinolinic acid and NAD, involved in various neurological pathways and reactions. It is known that quinolinic acid is found in amounts higher than required, in various neurodegenerative diseases such as Alzheimer’s, Huntington’s, etc, which can be a cause or effect relationship. From various studies going on in this respect, we found an intermediary enzyme 3-hydroxyanthranilic acid dioxygenase, which is an alpha-ketoglutarate-dependent dioxygenase, and uses Fe(II) as a cofactor. In this study, we report iron dependence of this enzyme in conversion of its substrate 2-hydroxyanthranilate into 2-amino-3-carboxymuconic semialdehyde, which is also an immediate upstream product to Quinolinic acid, thus linking Bna1 role in neurodegenerative diseases.
